Abstract

The combination of 0.1 mg/1 NAA and 2.5 mg/l BA stimulated lateral buds of Cynara Scolymus to form callus after 4-weeks and regenerated shoot form the callus in 8 weeks. This combination stimulated shoot-tips, forming multiplication shoots after 8 weeks. These different results were due to endogenous such as Auxin, Cytokinin, and ABA level in the lateral buds (higher than in the shoot-tips) while the Giberelin level in lateral buds was lower than the shoot-tip. The shoot was planted on the MS medium with the combination of 5 mg/l BA for forining 308 shoot per year. The combination of 0,5 mg/l GA3 and 0,5 mg/1 BA stimulated meristem to form shoot. Flower-head form shoot on the 1 mg/l TDZ medium. Shoot-tips and thin cell layer of shoot- tips formed callus when the medium occurred the combination of Auxin and Cytokinin. They were incubated in growth chamber at 222 °C with 16h/ daylight form inflorescent lamps at 2000 lux. The combination of 15 mg/l acid Ascorbic retarded browning process of the explain in in-vitro culture. The treatment of 100mg/1 BA stimulated the development of lateral buds.
